[Abstract]

Objective:The death rate of ovarian cancer is the highest in the Gynecology tumor.The difficulty to cure ovarian cancer is correlated with unclear symptom during early stage and metastasis during later stage,as well as unlear mechanism of generation and development.So it is the vital important to know the biologic activity clearly and evaluate the state of its generation and development.It is well known that apoptosis and angiogenesis play an important role in the process of generation and development of ovarian cancer.Many genic proteins about apoptosis and angiogenesis have been discoveried recently,such as:p53, p21, bcl-2, c-myc, VEGF, MVD etc.Now new-discoveried genic protein ?-survivin and HIF-1? become the issue to study carcinoma.To indentify survivin and HIF-1? in the mechanism of generation and development of ovarian cancer and relationship with apoptosis and angiogenesis in carcinoma,we investigate the expression of survivin and HIF-1? in ovarian cancer, ovarian benign tumor and ovarian normal tissue as well as the relationship with stage, grade, histological type of ovarian cancer.Materials and methods:(1) The expression of survivin and HIF-1? protein was studied by Flow Cytometry and Immunfluorescense in<WP=8>49 cases of ovarian cancer ,32 cases of benign tumor,and 13 cases of normal tissues for controls.FIGO stage?-? were retrieved from our files, including 11cases of stage ?,17cases of ?, 19cases of?,and 2 cases of ?.The mean age of patients was 46.5?8.5 years,and the mean age of 13 normal tissues was 44?8.2 years.(2)Every specimen was divided into two samples.One sample was fixed in nautral-buffered 4% Formalin and embedded in paraffin and then disagnoesd by the pathologist.The other sample was fixed in 70% alcohol and to be used to detect apoptosis rate.We took fluorescence index as the norm of survivin and HIF-1? protein quantitative expression.Results:(1) Expression of survivin, HIF-1?and apoptosis rate in different ovarian tissue:The expression of survivin and HIF-1?are different respectively,especial in ovarian cancer, P<0.01.The expression in ovarian cancer are both higher than that of benign and normal tissues, P<0.01.The apoptosis rates are different in different ovarian tissues, P<0.01, and the comparision of each other is different significantly,P<0.01.(2)The expressin of survivin and HIF-1?in different stage,grade,histological type of ovarian cancer:The expression of HIF-1?in the early stage of ovarian cancer is much higher than that of later stage , P<0.05. There are no significances among the expression of HIF-1? in different Grade and histological type.The expression of survivin in cervical endometrial-like adenocarcinoma is much higher than that of teratolarconoma and dysgerminoma P<0.05, but no difference with serous adecarcinoma and mucinous adenocarcinoma. The expression of survivin is no correlation with different grade and type, P>0.05.(3)The correlation coefficient was<WP=9>r=0.734 between survivin and HIF-1?,r=-0.479 between survivin and AP, r=0.753 between HIF-1? and VEGF in ovarian tumor, all P<0.01.Conclusions: The quantitative expression of survivin and HIF-1?in ovarian cancer are much higher than in benign tumor and normal tissue, and much correlated with apoptosis rate and the expression of VEGF. These demonstrate that survivin and HIF-1?are much related to the apoptosis and angiogenesis of ovarian tumor, as well as play an very important role in the mechanism of generation and development of ovarian tumor. Survivin and HIF-1? may be to become the promising tumor marker and aim of gene treatment.
